Resume

Tuberculosis remains a worldwide major public health problem. BCG, the vaccine which is used to prevent the disease has an efficacy that varies considerably. New tuberculosis vaccines have been developed. The evaluation of their efficacy is to carry out and might take several years. Surrogate markers of immunological protection are necessary to evaluate human vaccine trials. We have studied these markers by testing peripheral blood cells mRNA expression. Expression of TNF-receptors, TNFR1 and TNFR2 on one hand, and apoptotic proteins, FLICE and FLIPs on the other hand were compared between cohorts. These cohorts involved 12 tuberculosis patients, their household contacts (n=40) and 4 controls. Moreover, expression between infected and uninfected persons was compared. mRNA was quantified by Real-time Reverse Transcription-Polymerase Chain Reaction. The results suggest that TNFR1 and TNFR2 could be associated with the protection against TB, likewise FLICE could be associated with immunity against M. tuberculosis and FLIPs could be linked to infection with the bacteria. However, more cohorts of subjects, as well as other markers of anti-tuberculosis immunology should be studied
